Behavior and peripheral amine concentrations in relation to ractopamine feeding, sex, and social rank of finishing pigs

Aggression can impair productivity and well-being. The association between aggression in finishing pigs and the feed additive ractopamine (RAC), a β-adrenoreceptor agonist, is unknown and warrants further investigation. Our goal was to examine behavioral activity, including aggression, in the home pen and concentrations of peripheral amines in barrows and gilts, taking into account diet (RAC) and social rank. Sixty-four finishing pigs, housed in pens of 4 by sex, were fed either a control (CTL) or RAC-added (5 mg/ kg for 2 wk plus 10 mg/kg for another 2 wk) diet. The top dominant and bottom subordinate pigs in each pen were determined at mixing (2 wk pretrial). The behavior of all pigs was recorded continuously during the pretrial week (baseline) and for the following 4 wk. These behavioral data were used to evaluate home pen aggression, including the number of agonistic interactions (AINX) and constituent aggressive actions, during a 3-h period (0800 to 1100 h) once per week and their change in relation to the baseline. Time-budget behaviors and postures were analyzed over eight 24-h periods (2 d/wk) using 10-min instantaneous scan sampling that focused on only the dominant and subordinate pigs in each pen. These 2 pigs were also subjected to blood collection once per week during the trial to determine concentrations of dopamine, norepinephrine, epinephrine, and serotonin (5-HT) using HPLC. Gilts performed more bites and total actions per AINX than barrows, and RAC-fed gilts increased bites and pursuits, whereas these behaviors decreased compared with baseline values in all other subgroups. Gilts fed RAC increased the total number actions per AINX, whereas the occurrence of AINX decreased for all subgroups. Overall, RAC-fed pigs were more behaviorally active, spending more time alert, bar biting, and sham chewing compared with CTL pigs. The dominant RAC-fed pigs tended to have the greatest norepinephrine concentrations among the tested subgroups. Dominant barrows had greater epinephrine concentrations than subordinate barrows. The RAC-fed gilts tended to have lesser 5-HT concentrations than CTL gilts, whereas concentrations were similar in barrows. Greater activity and the increase in oral-related behaviors observed in RAC-fed pigs may be mediated by the increase in arousal caused by RAC. Intensified aggression in gilts, especially when fed RAC, may be linked to reduced central 5-HT and greater noradrenergic activity, and further research on brain neurotransmitters in gilts is needed.

Aggressiveness and brain amine concentration in dominant and subordinate finishing pigs fed the β-adrenoreceptor agonist ractopamine

Under farm conditions, aggression related to the formation of social hierarchy and competition for resources can be a major problem because of associated injuries, social stress, and carcass losses. Any factor that may affect the regulation and amount of aggression within a farmed system, for instance, feeding the β-adrenoreceptor agonist ractopamine (RAC), is therefore worthy of investigation. The objectives of this study were to assess the effects of the widely used swine feed additive RAC, considering also the effects of sex and social rank on aggressiveness and concentrations of brain amines, neurotransmitters essential for controlling aggression, in finishing pigs. Thirty-two barrows and 32 gilts (4 pigs/pen by sex) were fed either a control diet or a diet with RAC (Paylean, Elanco Animal Health, Greenfield, IN) added (5 mg/kg for 2 wk, followed by 10 mg/kg for 2 wk). The top dominant and bottom subordinate pigs (16 pigs/sex) in each pen were determined after mixing by a 36-h period of continuous behavioral observation. These pigs were then subjected to resident-intruder tests (maximum 300 s) during the feeding trial to measure aggressiveness. At the end of wk 4, the amygdala, frontal cortex, hypothalamus, and raphe nuclei were dissected and analyzed for concentrations of dopamine (DA); serotonin (5-HT); their metabolites 3,4-dihydroxyphenyl acetic acid (DOPAC) and homovanillic acid, and 5-hydroxyindoleacetic acid (5-HIAA), respectively; norepinephrine; and epinephrine using HPLC. Ractopamine-fed gilts performed more attacks during the first 30 s of testing than pigs in all other subgroups. By the end of the resident-intruder test (300 s), the dominant control gilts and barrows, and both dominant and subordinate RAC-fed gilts performed the greatest percentage of attacks. Gilts had decreased norepinephrine and DOPAC concentrations in the amygdala and frontal cortex, and when fed RAC, gilts also had the least 5-HIAA concentration and greatest DA turnover rate in the amygdala. The 5-HT concentration was less in the frontal cortex of gilts compared with barrows and in the raphe nuclei (single site for brain 5-HT synthesis) of dominant gilts. Ractopamine may be affecting aggressive behavior through indirect action on central regulatory mechanisms such as the DA system. The aggressive pattern observed in the tested pigs, especially in gilts, is likely linked to brain monoamine profiling of a deficient serotonergic system in the raphe nuclei, amygdala, and frontal cortex, and enhanced DA metabolism in the amygdala, brain areas vital for aggression regulation.

Observations of market pigs following transport to a packing plant

A field study was conducted to record observations of 41,744 market-weight pigs upon arrival at a commercial abattoir to evaluate the relationships between various independent factors and open-mouth breathing, skin discoloration, lameness, unloading time, and mortality during transport. Observations were recorded from 242 trailer loads on 46 separate days over a period of 14 mo. Travel time (<2.5 h or ≥2.5 h), wait time before unloading (<20 min or ≥20 min), loading pressure (<260 kg of BW/m2 trailer floor space or ≥260 kg of BW/m2 trailer floor space), ambient temperature while unloading (<17°C or ≥17°C), and trailer type [potbelly (PB); straight-deck with conventional unloading doors (SDC); or straight-deck with wide unloading doors (SDW)] were recorded for each load. Open-mouth breathing was more prevalent in pigs when transported on PB trailers compared with that of SDC or SDW trailers, and at warmer temperatures (≥17°C). Skin discoloration was more prevalent among pigs unloaded at temperatures ≥17°C. Lameness was more prevalent after shorter travel times at greater loading pressure compared with shorter travel times at decreased loading pressure. Unloading time for PB trailers was longer than for SDC and SDW. Mortality rates during transport were minimal (0.06%) in the deliveries that we observed, and there were no significant relationships between mortality and any independent variable tested. Wait time before unloading was not associated with any of the dependent variables included in the statistical model. In conclusion, warmer ambient temperatures (≥17°C) and the use of PB trailers are associated with an increased incidence of open-mouth breathing and skin discoloration, and longer unloading times after the transport of market pigs.

Effects of different moving devices at loading on stress response and meat quality in pigs

Although there is increasing evidence regarding the negative welfare and meat quality implications of electric prod use for slaughter-weight pigs, this handling tool continues to be used. Therefore, the behavioral and physiological response and carcass and meat quality of 360 pigs being loaded onto a truck for transportation to slaughter according to 3 handling procedures were studied. The 3 handling procedures were 1) moving with an electric prod and board from the finishing pen to the truck (EP); 2) moving with a board and a paddle from the finishing pen to the truck (PAD); 3) moving with a board and a paddle from the finishing pen and using a compressed air prod in the ramp before going into the truck (CAP). A subpopulation of 144 pigs (48 pigs/treatment) was equipped with heart rate monitors. Blood samples were collected from the same animals at exsanguination for the analysis of creatine phosphokinase and lactate. Data were analyzed using an ANOVA for factorial design, with the animal as the experimental unit. Behavior was analyzed with MIXED model procedure with treatment as a fixed effect. During loading, EP pigs slipped and fell and overlapped more often, but stopped and attempted turns less often than CAP or PAD. With CAP, pigs made more 180° turns than with PAD or EP. Loading with EP led to more and longer vocalizations than loading with CAP or PAD. Loading took longest with CAP and was quickest with EP. Pigs handled with EP had a greater heart rate than those moved with PAD and CAP at loading, wait at loading, at unloading, and in lairage. Pigs loaded with EP had greater lactate concentrations in blood at exsanguination compared with pigs handled with CAP, with pigs loaded with PAD being intermediate. Furthermore, ultimate pH values in the semimembranosus and adductor muscles of EP pigs were greater compared with those from PAD and CAP pigs. Greater incidence of blood-splashed hams was found in EP pigs compared with PAD and CAP pigs. Therefore, considering animal welfare, carcass bruising, and blood splashes standpoints, EP should be replaced with PAD or CAP. However, additional research is necessary to identify methods that improve the loading efficiencies of PAD and CAP without adversely affecting animal welfare parameters.

A single nucleotide polymorphism set for paternal identification to reduce the costs of trait recording in commercial pig breeding

In animal breeding, recording of correct pedigrees is essential to achieve genetic progress. Markers on DNA are useful to verify the on-farm pedigree records (parental verification) but can also be used to assign parents retrospectively (parental identification). This approach could reduce the costs of recording for traits with low incidence, such as those related to diseases or mortality. In this study, SNP were used to assign the true sires of 368 purebred animals from a Duroc-based sire line and 140 crossbred offspring from a commercial pig population. Some of the sires were closely related. There were 3 full sibs and 17 half sibs among the true fathers and 4 full sibs and 35 half sibs among all putative fathers. To define the number of SNP necessary, 5 SNP panels (40, 60, 80, 100, and 120 SNP) were assembled from the Illumina PorcineSNP60 Beadchip (Illumina, San Diego, CA) base on minor allele frequency (>0.3), high genotyping call rate (≥90%), and equal spacing across the genome. For paternal identification considering only the 66 true sires in the data set, 60 SNP resulted in 100% correct assignment of the sire. By including additional putative sires (n = 304), 80 SNP were sufficient for 100% correct assignment of the sire. The following criteria were derived to identify the correct sire for the current data set: the logarithm of odds (LOD) score for assigning the correct sire was ≥5, the number of mismatches was ≤1, and the difference in the LOD score between the first and the second most likely sire was >5. If the correct sire was not present among all putative sires, the mean LOD for the most likely sire was close to zero or negative when using 100 SNP. More SNP would be needed for paternal identification if the number of putative sires increased and the degree of relatedness was greater than in the data set used here. The threshold for the number of mismatches can be adjusted according to the practical situation to account for the tradeoff between false negatives and false positives. The latter can be avoided efficiently, ensuring that the correct father is being sampled. Nevertheless, a restriction on the number of putative sires is advisable to reduce the risk of assigning close relatives.

Effect of prenatal stress on subsequent response to mixing stress and a lipopolysaccharide challenge in pigs

Sows subjected to prenatal stress have been found to produce offspring that have altered responses to stress. Our objective was to determine if exposing a sow to stress would alter the response of the offspring to lipopolysaccharide (LPS) at 2 mo of age or their response to mixing stress at 4 mo of age. Sow treatments consisted of intravenous injections of ACTH (1 IU/kg of BW), exposure to rough handling for a 10- min duration (rough), or no treatment (control) once per week from d 42 to 77 of gestation. At 2 mo of age, pigs from each treatment, 1 per litter (n = 21, 17, and 15 for the ACTH, rough, and control treatments, respectively), were challenged with 2 μg of LPS/kg of BW or saline, or served as a noninjected control. Their behavioral response to a human approach test and salivary cortisol were measured. At 4 mo of age, 1 pig from each treatment (n = 14, 14, and 15 for the ACTH, rough, and control treatments, respectively) was taken from its home pen and placed in a pen of unfamiliar pigs. At this time, a punch biopsy wound (6 × 6 mm) was created to measure the ability of the pig to heal the wound. At this same time, each pig received a 1-mL intramuscular injection of 20% ovine red blood cells (oRBC), and then a second injection of oRBC at 21 d postmixing. Blood samples were collected 3 times per week for 2 wk and then once a week for 4 more weeks. Blood samples were analyzed for cortisol, porcine corticosteroid- binding globulin, antibody response to oRBC, and nitric oxide production by macrophages. Behavior was recorded during the first 5 d after mixing. All pigs in the LPS challenge responded with characteristic sickness behavior; however, pigs in the rough treatment showed less sickness behavior than those in the other 2 treatments. Maternal stress treatment did not affect salivary cortisol. Pigs from all treatments responded similarly to mixing stress with regard to cortisol, porcine corticosteroid-binding globulin, antibody titers, nitric oxide production, and hematology measures, and all pigs experienced the same amount of aggression in response to mixing. Without altering peripheral measures of stress responsivity, prenatal stress enhanced the ability of pigs to cope with a simulated immune challenge, which could prove to be an adaptation to challenging environments.

Effects of tryptophan supplementation on aggression among group-housed gestating sows

The objective of this study was to determine the effects of dietary Trp supplementation on mixing-induced aggression and the associated stress, and on reproductive performance in gestating sows. After weaning, sows were mixed in pens with electronic sow feeders on concrete-slatted floors. Each pen housed 21 sows, with approximately 2.7m2/sow of floor space allowance. Multiparous sows (n = 168) from 8 breeding groups were used, with 4 groups assigned to a control diet and 4 groups assigned to a high-Trp diet. Control sows received corn- and soybean meal-based diets throughout gestation (0.15% Trp) and lactation (0.21% Trp). Three days before and after mixing, sows assigned to the high-Trp treatment received approximately 2.3 times the dietary Trp (0.35% in the gestation diet and 0.48% in the lactation diet) fed to control sows. Six focal sows (2 sows from each of parity 1, 2, and 3 or greater) in each pen were designated and videotaped for 72 h after mixing to determine the type and number of aggressive interactions among sows. Before and 48 h after mixing, saliva samples were collected from focal sows, and scratches were assessed on all sows. Data were analyzed using the FREQ and GLIMMIX procedures (SAS Inst. Inc., Cary, NC). Aggression among sows was intense during the initial 6 h and decreased between 6 and 72 h after mixing. The initial aggression caused scratches and increased cortisol concentrations. Mature sows tended to fight for longer periods (112 vs. 52 s/h per sow) but had fewer scratches caused by aggression (injury score = 4.3 vs. 6.5) than parity-1 sows. Supplementation of dietary Trp reduced the total duration of head-to-head knocking but did not affect other aggressive behaviors. There was no difference between dietary treatments in injury scores or saliva cortisol concentrations. Sows in the high-Trp treatment had more total piglets born (12.5 vs. 10.5 pigs/litter) and more stillborn piglets (1.5 vs. 0.8 pigs/litter), but had no significant change in piglets born alive (10.8 vs. 9.7 pigs/litter) compared with control sows. The results indicate that the initial aggression after mixing caused more injuries in young sows than in mature sows. Supplementation of dietary Trp at 2.3 times the control amount for a short period did not effectively reduce aggression and the associated stress in sows at mixing.

Metabolizable energy content of refined glycerin and its effects on growth performance and carcass and pork quality characteristics of finishing pigs

Two studies were conducted with refined glycerin (97.7 and 97.5% glycerin for studies I and II, respectively) to determine ME content and effects on growth performance and carcass and pork quality measures of finishing pigs. An energy balance study using 24 barrows (21.5kg initial BW) determined the apparent ME content of glycerin using a generalized randomized block design with 2 dietary treatments: 1) control (99.85% corn + vitamins and minerals) and 2) glycerin (30% of corn in the control diet replaced with glycerin). A 7-d adaptation was followed by a 5-d collection period for feces and urine. The energy content of diets, feces, and urine was determined by bomb calorimetry. The DE of the glycerin diet was greater than that of the control diet (4,298 vs. 3,902 kcal/kg of DM); however, the ME content of the 2 diets was similar (3,820 vs. 3,723 kcal/kg of DM). The ME of refined glycerin (estimated by difference) was 3,584 kcal/ kg of DM. A growth study was conducted with 128 gilts housed in groups of 4 and reared from 92.5 kg of BW for a 28-d period, using a split-plot design with a 4 × 2 factorial arrangement of treatments: 1) dietary glycerin level (0, 5, 10, and 15%) and 2) preslaughter handling (gentle vs. intense). The handling treatment was included to simulate the range in handling intensities that are likely to be experienced in practice. At the end of the 28-d period, one-half of the pens on study were slaughtered and used for carcass and pork quality evaluation with 2 pigs from each pen being subjected to each of the preslaughter handling treatments. There were no interactions between dietary glycerin and preslaughter handling treatment. Dietary glycerin had no effect on growth performance, carcass measures, or meat quality. There were no differences between the gentle and intense handling treatments for carcass or pork quality measures. In conclusion, feeding glycerin to finishing pigs at up to 15% of the diet had no negative effect on growth performance or carcass and pork quality characteristics.

Relative bioavailability of phosphorus in inorganic phosphorus sources fed to growing pigs

The relative bioavailability of P in 5 sources of inorganic P was determined using growing pigs. The 5 sources of inorganic P were dicalcium phosphate (DCP), monocalcium phosphate (MCP) containing 50% MCP (MCP50), MCP containing 70% MCP (MCP70), MCP containing 100% MCP (MCP100), and monosodium phosphate (MSP). A total of 11 diets were formulated. The basal diet was formulated to contain 0.10% P, and 10 additional diets were formulated by adding 0.07 or 0.14% P from each of the 5 P sources to the basal diet. Growing pigs (n = 44; initial BW: 16.8 kg) were individually housed and randomly allotted to the 11 experimental diets. Feed was provided on an ad libitum basis throughout the 28-d experimental period. At the conclusion of the experiment, all pigs were killed, and 4 bones (i.e., the third and fourth metacarpals on both front feet) were harvested. Bone-breaking strength, bone ash, and Ca and P concentrations were determined. The concentration of bone ash increased as MCP50, MCP70, MCP100, or MSP were added to the basal diet, and the concentration of bone P also increased as MCP70, MCP100, or MSP were added to the basal diet. The relative bioavailability of P in each of the feed phosphates was determined using slope ratio methodologies based on breaking strength, and expressed relative to MSP. The slope of the regression line for diets containing MSP or MCP100 was steeper than the slope for pigs fed the diet containing DCP, but not different from that of pigs fed diets supplemented with MCP50 or MCP70. In conclusion, P in MSP and MCP100 is more bioavailable than P in DCP, but there were no differences within MCP sources.

Commercially produced spray-dried porcine plasma contains increased concentrations of porcine circovirus type 2 DNA but does not transmit porcine circovirus type 2 when fed to naive pigs

The porcine circovirus type 2 (PCV2) antibody and DNA status of porcine plasma products collected during the commercial spray-drying process were evaluated. Samples evaluated included 52 pooled liquid plasma (fresh) samples collected at 14 regional abattoirs before transport to 1 of 2 spray-drying facilities, 32 pooled liquid plasma (concentrated) samples collected after arrival at the spray-drying facilities at different stages before the spray-drying process, and 32 samples in powdered form (spray-dried) collected after spray drying. All 116 samples were positive for PCV2 antibody, with PCV2 ELISA sample-to-positive ratios ranging from 9.2 to 13.6 on a DM basis. Porcine circovirus type 2 DNA (4.5 to 7.9 log10 PCV2 copies/mL, DM basis) was present in 82.7% (43/52) of the fresh plasma samples, 71.9% (23/32) of the concentrated plasma samples and 78.1% (25/32) of the spray-dried plasma samples, with a greater prevalence of PCV2b than PCV2a. To determine the infectivity of PCV2 DNA-positive commercial spray-dried plasma, nine 10-wk-old 68-kg PCV2-naive pigs were randomly assigned to 1 of 3 treatment groups and rooms: 1) a negative control (no plasma in the feed, not inoculated with PCV2); 2) a positive control (no plasma in the feed, inoculated with PCV2); and 3) plasma-fed pigs (4% porcine plasma in the feed for 42 d, not inoculated with PCV2). All positive control pigs became viremic by 7 d postinoculation and seroconverted by 42 d postinoculation, whereas pigs in the negative control group and in the spray-dried plasma group were PCV2 PCR negative and did not seroconvert to PCV2 for the duration of the study. The results indicate that PCV2 DNA and antibodies are commonly found in commercial spray dried plasma. However, no evidence of infectivity of the PCV2 DNA was found in naive pigs when commercial spray-dried plasma was included in the diet under the conditions of this study.
