Immunoblot analysis with polyclonal anti-pIFN-g-antibody\u00a0demonstrated that the recombinant porcine IFN-g (rpIFN-g) protein was expressed in the L. lactis transformants as a secreted\u00a0product. Activity of rpIFN-g was confirmed by ability to upregulate class II major histocompatibility complex (MHC) on cells of\u00a0the porcine monocytic cell line 3D4\/31. The L. lactis producing biologically active rpIFN-g will be used to investigate its\u00a0possible ability to modulate the allergic immune response phenotype of pigs. \u00a0Biologically active rpIFN-g was produced in L.\u00a0lactis and upregulated MHC class II expression on\u00a0porcine monocytic cells suggesting that the modified\u00a0L. lactis might be able to enhance type 1 immune\u00a0response and reduce susceptibility to allergy in\u00a0animals\/humans. The researchers have\u00a0engineered and expressed afunctional rpIFN-g gene in L. lactis that would\u00a0allow delivery to the pig\u2019s intestinal mucosa. \u00a0Although use of this construct is limited by lack\u00a0of control over quantity and conditional expression\u00a0of recombinant protein production, containment of\u00a0live genetically modified L. lactis secreting human\u00a0IL-10 in pigs is possible using an auxotrophic\u00a0thymidine mutant (Steidler et al., 2003).
